Human Plasma a , - Macroglobulin and von Willebrand Factor Possess Covalently Linked ABO ( H ) Blood Group Antigens in Subjects With Corresponding A B 0 Phenotype

چکیده
We recently identified ABO(H) blood group structures in Asn-linked sugar chains of human von Willebrand factor (vWF) purified from factor Vlll concentrates (J Biol Chem 267:8723, 1992). We surveyed plasma glycoproteins carrying ABO(H) blood group antigens by Western blotting analysis and sandwich enzyme-linked immunosorbent assay using blood group-specific monoclonal antibodies (MoAbs) and a lectin. Two major plasma proteins showing apparent molecular weight of about 180 Kd and 270 Kd by sodium dodecyl sulfate polyacrylamide gel electrophoresis reacted with blood group-specific MoAbs and Ulex europaeus lectin I in accordance with donor blood group. Direct sequence analysis of the protein bands showed their identity with the N-terminal sequences of a,-macroglobulin (a,M) and vWF, respectively. The two bands also

von Willebrand factor (vWF) is synthesized exclusively by endothelial cells and megakaryocytes, and stored in the intracellular granules or constitutively secreted into plasma. ABO blood group antigens are covalently associated with asparagine-linked sugar chains of plasma vWF. Although he devoted himself to research in bacteriology, haematology and immunology, he tried to maintain a close relationship with clinical medicine. Landsteiner’s intellectual curiosity was centred on fundame...
متن کاملThe effect of ABO blood group on the diagnosis of von Willebrand disease.
In order to firmly establish a normal range for von Willebrand factor antigen (vWF:Ag), we determined plasma vWF:Ag concentrations in 1,117 volunteer blood donors by quantitative immunoelectrophoresis. The presence of the ABO blood group has a significant influence on vWF:Ag values; individuals with blood group O had the lowest mean vWF:Ag level (74.8 U/dL), followed by group A (105.9 U/dL), th...

Introduction The antigens of the ABO blood group system (A, B and H determinants, respectively) are complex carbohydrate molecules on the extracellular surface of red blood cells1. However, besides being expressed on red blood cells, ABO antigens are also strongly expressed on the surface of a variety of human cells and tissues, including the epithelium, sensory neurones, platelets, and the vas...
